Effect of Anti - lL - 3 Antibody on TC - 1 CM Stimulated

چکیده
We reported previously that a cell line (TC-1 ) derived from adherent marrow cells produced colony-stimulating factor 1 (CSF-1 ) and a separate activity that acts synergistically with CSF-1 to stimulate giant macrophage colonies. We now report that an activity in TC-1 conditioned media (CM) separate from CSF-1 also synergizes multilineage colony formation by pure interleukin 3 (IL 3) and a crude source of granulocyte-macrophage colony-stimulating activity (GMCSA) (murine lung-conditioned media). IL 3-induced megakaryocyte colony formation is also synergized. The CSF-1 -dependent synergistic activity is not blocked by antibodies to IL 3 and is characterized as a nondialyzable (mol wt cutoff 3.000). heat-stable (56 ‘C. 30’) activity that binds to DE-52 cellulose under conditions in which IL 3 does

T Cells From Eosinophilic Patients Produce Interleukin - 5 With tnterleukin - 2 Stimulation
Anti-murine (m) interleukin-5 (IL-5) antibody was found to inhibit eosinophil (Eo) colony formation stimulated by recombinant human (rh) IL-5, but did not inhibit the production of Eo stimulated by rh lL-3 or granulocyte-macrophage colony-stimulating factor (GM-CSF). Conditioned medium (CM) prepared from eosinophilic patients’ T cells with interleukin-2 (IL-2) stimulation (T-lL-2-CM), was found...
متن کاملSynergistic Anti-Inflammatory Activity of the Antimicrobial Peptides Human Beta-Defensin-3 (hBD-3) and Cathelicidin (LL-37) in a Three-Dimensional Co-Culture Model of Gingival Epithelial Cells and Fibroblasts
Given the spread of antibiotic resistance in bacterial pathogens, antimicrobial peptides that can also modulate the immune response may be a novel approach for effectively controlling periodontal infections. In the present study, we used a three-dimensional (3D) co-culture model of gingival epithelial cells and fibroblasts stimulated with Aggregatibacter actinomycetemcomitans lipopolysaccharide...
متن کاملTopical Treatment of Rosacea with Ivermectin Inhibits Gene Expression of Cathelicidin Innate Immune Mediators, LL-37 and KLK5, in Reconstructed and Ex Vivo Skin Models
INTRODUCTION Numerous intrinsic and extrinsic factors have been associated with the pathophysiology of rosacea, including dysregulation of innate immunity. A high level of cathelicidin antimicrobial peptides (e.g., LL-37) has been shown in the facial skin of patients with rosacea. Excessive production of both LL-37 and KLK5, the serine protease responsible for its cleavage, has been suggested t...
متن کاملInsulin activates glycogen synthase by a novel PI 3-kinase/p70s6k dependent pathway in 3T3-L1 adipocytes.
Insulin causes the activation of a tyrosine kinase activity in the intracellular domain of its receptor, the major substrate of this kinase being the multifunctional docking protein E l . Phosphorylation of specific tyrosine residues on IRSl allows this protein to interact with and activate a number of downstream signalling molecules including phosphoinositide 3-kinase (PI 3-kinase), SHPTP2 and...
متن کاملDual combination therapy targeting DR5 and EMMPRIN in pancreatic adenocarcinoma.
The goal of the study was to assess the efficacy of combined extracellular matrix metalloprotease inducer (EMMPRIN)- and death receptor 5 (DR5)-targeted therapy for pancreatic adenocarcinoma in orthotopic mouse models with multimodal imaging. Cytotoxicity of anti-EMMPRIN antibody and anti-DR5 antibody (TRA-8) in MIA PaCa-2 and PANC-1 cell lines was measured by ATPlite assay in vitro. The distri...
